Research

Her research project aims at understanding the impacts of polyunsaturated fatty acids (PUFAs), oxylipins and lipophilic micronutrients on vascular function. The approaches used to adress this question includes omics (transcriptomics, proteomics and lipidomics), human and animal intervention studies as well as in vitro experiments in immune and endothelial cells.

The term oxylipins refers to a superclass of lipid metabolites produced from oxygenation of polyunsaturated fatty acids (PUFAs). They include the prostanoids and thromboids produced by cyclooxygenases (COXs); leukotrienes, resolvins, hepoxilins and mid-chain alcohols generated by lipoxygenases (LOXs), epoxides and omega-terminal alcohols produced by cytochrome p450s (CYPs) as well as iso/neuroprostanes issued from the free radical mediated oxidation of PUFAs (Buczynski, 2009). Oxylipins play important roles in many physiological processes, such as blood clotting, blood vessel permeability, inflammation and immune cell behavior and several lines of evidence suggest that metabolic profiling of oxylipins could be a good reflection of vascular health. To develop and validate the use of metabolic profiling of oxylipin as an assessment of the vascular function, we are collaborating with the the IBMM laboratory (UMR 5247, CNRS), the MetaToul-Lipidomic plateau (Toulouse) and the West Coast Metabolomic Center, CA, USA (JW Newman). Notably, one objective is to enlarge the active target lists for oxylipin metabolite profiling, particularly with the non-enzymatic species such as neuroprostanes, which have been recently identified as new putative important contributors of vascular health (Gladine et al., PlosOne, 2014). By combining metabolomics of oxylipins with more targeted approches of molecular and cellular biology, we will aslo investigate the contribution of different classes of oxylipins in the cellular responses of immune and endothelial cells and their nutritional modulation.

Curriculum Vitae

Cécile Gladine obtained her PhD in Nutrition & Biochemistry in November 2006, after which she was recruited as a researcher at INRA, UMR 1019 Human Nutrition in 2007. From 2008 to 2009, she was appointed as a post-doctoral research fellow at the Food, Nutrition and Genomic group, AgResearch Research Institute (New-Zealand) and then return to the UMR 1019 in Clermont-Ferrand (France) where she has joined the Micronutrients and Cardiovascular team.
