Solving the problems related with aging is an important issue in human nutrition research. According to INSEE, in 2010, France had 5.7 million people over 75 years of age. This figure is expected to double by 2060. In addition to the specific pathologies of old age, during aging there is a decrease in the body's ability to cope with stresses. One of the factors that explains this phenomenon is the reduction in the mass of muscle proteins occurring from the third decade in humans.

The main function of muscle remains locomotion. This function may decline as a result of pathologies or accidents, but will inevitably deteriorate during aging. This phenomenon is self-sustaining because physical inactivity increases the loss of muscle mass that increases physical inactivity! The health of muscle is closely correlated with that of the locomotor system (bone, cartilage), and any alteration can have an impact on muscle mass. Muscle is also important in relation with energy expenditure. This is related to the amount of muscle mass, because of the energy costs of the renewal of muscle proteins. In addition, physical activity induces a very significant increase in energy expenditure. Furthermore, muscle is a site of oxidation of fatty acids and glucose. There will therefore be close relationships between muscle mass, adiposity, and insulin resistance. Finally, muscle is an emergency store of amino acids for the body. In the event of nutritional, infectious or traumatic stress, it is in the muscle that the body will find the amino acids necessary to cope with this stress. In total, a loss of muscle mass will reduce the body's ability to restore a state of good health, and may increase the morbidity and mortality of the subjects in case of serious pathology. It is therefore important to maintain muscle mass during aging.

My research theme is the study of the mechanisms of the age-related muscle protein loss. The aim is to propose strategies to slow it down in order to maintain people's autonomy and good health as long as possible.

I first analyzed the age-related evolution of the activities of synthesis and degradation of muscular proteins because they determine the evolution of muscle mass. I then analyzed the impact of modulators of these activities, exercise and nutrition.

In particular, I studied the effect of protein intake, because proteins have a direct effect on muscle metabolism. I analyzed the impact of the nature and kinetics of protein intake (chrono-nutrition). I applied the knowledge gained on this protein chrono-nutrition in the context of aging to another context, the context of obesity. The aim was to limit lean body mass losses during energy restriction in young obese or overweight subjects. I also interested in the interaction between oxidative stress, inflammation and muscle wasting during aging. I measured the age-related change in muscle oxidative damage and free radicals production, and tested the impact of polyphenol and antioxidant supplementation on post-prandial muscle protein metabolism.
